Hormonal Relations in the Phototropic Response IV. Light-Induced Changes of Endogenous Auxins in the Coleoptile.

چکیده
Beside indoleacetic acid (IAA), 3 auxins were found by chromatographic resolution of acidic fractions of Avena and Zea coleoptile tips. One of these auxins, designated P, occurred at levels of activity approaching those of IAA. The other 2 auxins, termed F and M, occurred at lower levels of activity. When the auxins of the excised coleoptile tips were isolated immediately after equilateral or unilateral irradiation with blue light at first positive energies, the ratio of IAA to the other auxins increases. This rise is the result of a decrease in P and F, and probably an increase in IAA. Light did not affect materially the total auxin content. It is suggested that P and F might be associated with the basipetal transport inequalities of IAA in phototropism.P has been partially characterized. Its R(F) on chromatograms developed in ammoniacal isopropanol is about 0.65. It is converted to IAA in vitro by heat. The ultraviolet absorption spectrum of chromatographically resolved P also suggests an indolyl complex. P is not readily transported basipetally, and the slope of its relative concentration-response curve (Avena section test) is lower than that of IAA. P does not appear to be any of the chemically characterized native auxins.

Blue light regulates an auxin-induced K+-channel gene in the maize coleoptile.
Auxin redistribution along gravistimulated maize coleoptiles causes differential expression of the auxin-induced K+-channel gene ZMK1 (Zea mays K+ channel 1) and precedes the curvature response. To evaluate the role of ZMK1 during phototropism, we here investigated blue light-stimulated coleoptiles. Four hours of blue light stimulation resulted in phototropic bending (23 degrees ). Rotation on ...
متن کاملElectrical and Curvature Responses of the Avena Coleoptile to Unilateral Illumination.
Introduction One fact that is agreed upon in the literature regarding the mechanism of phototropism is that the growth curvature results from an unequal distribution of auxin. There are at least three hypotheses as to how unilateral illumination causes the unequal distribution: 1. Photoinactivation of auxin on the lighted side of the plant. 2. A differential change in the rate of synthesis of a...
متن کاملPhototropism and photoinhibition of basipolar transport of auxin in oat coleoptiles.
We have proposed that the lateral inequality of auxin associated with the phototropic response is a consequence of a light-induced impairment of basipolar transport of the hormone. If this is so, the dose-response curve for photoinhibition of auxin transport should resemble that for phototropism. Further, the wavelength dependencies of the transport and tropic response should also be similar. O...
متن کامل6-Kilodalton Plasma Membrane Protein Can Be Correlated with the First- and Second-Positive Phototropic Curvature of Oat CoIeoptiIes'
The possible correlation between blue light-dependent phosphorylation of a 11 6-kD protein and phototropic responses of etiolated oat (Avena sativa 1.) seedlings was tested by a micromethod for protein phosphorylation. Quantitation of the basipetal distribution of this protein showed that the in vitro 32P phosphorylation values declined exponentially from tip to node, with more than 50% of the ...
متن کاملGravitational compensation and the phototropic response of oat coleoptiles.
Avena seedlings were germinated and grown while continuously rotated on the horizontal axis of a clinostat. The coleoptiles of these gravity-compensated plants were phototropically more responsive than those of plants rotated on a vertical axis. When the plants were compensated after unilateral irradiation, phototropic curvature of the shoot progressed for the next 6 hours, with the rate of cur...
